How to maintain the HPLC autosampler instruments?

With enough time to balance the column every day, you get the most "compensation" in dealing with problems, and your column life will be longer! It must be done. The new column should be tested on your own liquid chromatograph before use. The test conditions and samples on the test report attached to the column are used to determine the column efficiency. Also, in future use, the column should be tested from time to time.

Method / Step:


1: Installation of the ferrule column (without pre-column)

1. Insert the card holder into the core

2. Insert the two clamping pieces into the groove of the core so that the core is higher than the jacket (see left)

3. Push the ferrule holder that has been placed over the core until it is higher than the jacket

4. Screw vials PP cap and the card holder together and tighten by hand

5. Then connect the other end of the column in the same order.

6. Connect to the liquid chromatograph, the PEEK connector can be hand-screwed; if it is a stainless steel joint, use a special wrench

2: Installation of the ferrule column (plus pre-column)

1. Insert the card holder into the core

2. Insert the two clamping pieces into the groove of the core so that the jacket is higher than the core (see left)

3. Push the ferrule holder that has been placed over the core until it is higher than the jacket

4. Put the "bullet" pre-column into the ferrule

5. Screw the cap and the card holder together and tighten by hand

6. Then connect the other end of the column in the same order.

7. Connect to the HPLC , the PEEK connector can be hand-screwed; if it is a stainless steel joint, use a special wrench

Replace the column filter and the glass wool filter (you can also repair the column)

3: Before taking out the filter and glass piece of the reversed-phase core, the HPLC column should be flushed with water and methanol/acetonitrile, and the head of the repair tool should also take a small amount of methanol/acetonitrile to avoid removing the filter. And the glass wool filter brings out the packing in the column.

1. Put 2 of the repair tools into the top of the core

2. Gently screw autosampler vials the 3 in the repair tool into the core of the sleeve 2 and turn it clockwise to tighten

3. Hold the core in one hand and gently pull outward 3 in the other hand to remove the filter from the top of the core.

4. Gently remove the glass wool under the filter and the contaminated packing with a small shovel

5. Wet the new filler with methanol, then fill it in the excavated area and flatten it

6. Attach a new glass wool filter (picture on the left) and press the glass wool into the top of the core with 4 in the repair tool.

7. Place the top of the core with 2, then refer to the (left) to place the filter

8. Press down, then remove 2, then use 4 to flatten the edges of the filter

Precautions:

Balanced HPLC Columns Reversed-phase HPLC columns are stored in acetonitrile/water after factory testing. Make sure that the mobile phase you are using is miscible with acetonitrile/water. Since the HPLC column may dry out during storage or transportation, the column should be equilibrated with 10-20 column volumes of methanol or acetonitrile before analyzing the sample with the mobile phase; if the mobile phase you are using contains buffer salts, Should pay attention to the "transition" with pure water.
