SPME Vials vs Standard Headspace Vials

Time:2026/10/19   Pageviews:0    Share:

SPME vials (Solid Phase Microextraction vials, often interchangeably called SPME headspace vials) are gas-tight glass containers specially manufactured for solid-phase microextraction sampling in GC / GC-MS. They accommodate SPME fiber penetration, maintain stable headspace equilibrium under heating, and prevent volatile loss or contamination during extraction.

SPME is a solvent-free sample preparation technique. The SPME fiber pierces the vial septum to extract analytes either from the headspace above the sample (HS-SPME) or by direct immersion into liquid sample (DI-SPME). After equilibrium, the fiber is retracted and desorbed in the GC inlet for detection.

Common Volumes & Dimensions

Core Design Features for SPME

  1. Gas-tight sealing: Maintain stable phase equilibrium during heating & extraction; prevent volatile leakage and air ingress.
  2. Precision neck & opening: Optimized inner neck diameter for SPME fiber access. Too small opening risks fiber breakage.
  3. Low-bleed glass & certified options: Certified vials with low background contamination for trace VOC analysis (food, environmental, fragrance).
  4. Heat resistance: Stable under incubation temperature (common 40–90 °C) for headspace equilibrium.

Typical Applications

The fifference between SPME Vials and Standard Headspace Vials.
Item SPME Vials Standard Headspace Vials
Main purpose HS-SPME / SPME Arrow, fiber penetration Static headspace, gas sampling by needle
Vial lip Thicker lip Normal thin lip
Recommended septum Thin septum (1.5mm), PTFE/silicone Thick septum (3.0mm), PTFE/silicone
Core benefit Low coring, protect SPME fiber, good seal after crimping High pressure & heat resistance for headspace oven
Compatibility CTC PAL, Gerstel SPME autosampler Agilent, Shimadzu standard headspace sampler

Q: Can I use 1.5mm septa on regular 20mm crimp headspace vials for SPME?
A: Not recommended. Regular headspace vials have a thinner lip. 1.5mm thin septa require SPME vials with thicker vial lip to achieve proper gas-tight crimp seal. Using thin septa on standard vials may cause leakage during heating, leading to inconsistent analytical results.
